Fluorescence lifetime based bioassays

چکیده
Fluorescence lifetime (FLT) is a robust intrinsic property andmaterial constant offluorescentmatter. Measuring this important physical indicator has evolved from a laboratory curiosity to a powerful and established technique for a variety of applications in drug discovery, medical diagnostics and basic biological research. This distinct trendwasmainly driven by improved andmeanwhile affordable laser and detection instrumentation on the one hand, and the development of suitable FLT probes and biological assays on the other. In this process two essential working approaches emerged. Thefirst one is primarily focused on high throughput applications employing biochemical in vitro assays with no requirement for high spatial resolution. The second evenmore dynamic trend is the significant expansion of assaymethods combining highly time and spatially resolved fluorescence data by fluorescence lifetime imaging. The latter approach is currently pursued to enable not only the investigation of immortal tumor cell lines, but also specific tissues or even organs in living animals. This review tries to give an actual overview about the current status of FLT based bioassays and the wide range of application opportunities in biomedical and life science areas. In addition, future trends of FLT technologies will be discussed.

متن کاملMetal-Enhanced Fluorescence of Phycobiliproteins from Heterogeneous Plasmonic Nanostructures.
We report here the use of plasmonic metal nanostructures in the form of silver island films (SiFs) to enhance the fluorescence emission of five different phycobiliproteins. Our findings clearly show that the phycobiliproteins display up to a 9-fold increase in fluorescence emission intensity, with a maximum 7-fold decrease in lifetime when they are assembled as a monolayer above SiFs, as compar...
متن کاملCarbostyril derivatives as antenna molecules for luminescent lanthanide chelates.
Luminescent lanthanide complexes consisting of a lanthanide-binding chelate and organic-based antenna molecule have unusual emission properties, including millisecond excited state lifetimes and sharply spiked spectra, compared to standard organic fluorophores. We have previously used carbostyril (cs124, 7-amino-4-methyl-2(1H)-quinolinone) as an antenna molecule (Li and Selvin, J. Am. Chem. Soc...
متن کاملSingle-molecule photon stamping FRET spectroscopy study of enzymatic conformational dynamics.
The fluorescence resonant energy transfer (FRET) from a donor to an acceptor via transition dipole-dipole interactions decreases the donor's fluorescent lifetime. The donor's fluorescent lifetime decreases as the FRET efficiency increases, following the equation: E(FRET) = 1 - τ(DA)/τ(D), where τ(D) and τ(DA) are the donor fluorescence lifetime without FRET and with FRET. Accordingly, the FR...
متن کاملLabel-free detection of single protein molecules using deep UV fluorescence lifetime microscopy.
We present the detection of single beta-galactosidase molecules from Escherichia coli (Ecbeta Gal) using deep UV laser-based fluorescence lifetime microscopy. The native fluorescence from intrinsic tryptophan emission has been observed after one-photon excitation at 266 nm. Applying the time-resolved single-photon counting method, we investigated the fluorescence lifetime distribution and the b...
